Blueprint Medicines Reports Positive 4-Year Data for AYVAKIT

Blueprint Medicines, a Sanofi company, announced new four-year data from its PIONEER study showing sustained benefit and long-term safety for its drug AYVAKIT in treating indolent systemic mastocytosis. A separate real-world study also highlighted the drug's effectiveness in improving symptoms of the rare disease.

Indolent systemic mastocytosis (ISM) is a rare, chronic blood disorder caused by an overproduction of mast cells, which can accumulate in various tissues. This buildup leads to a wide range of debilitating symptoms, including skin lesions, gastrointestinal problems, severe fatigue, and cognitive issues often described as "brain fog." AYVAKIT's active ingredient, avapritinib, is a kinase inhibitor that functions by targeting the primary underlying cause of the disease. In approximately 95% of cases, systemic mastocytosis is driven by a specific genetic mutation known as KIT D816V, which AYVAKIT is designed to potently and selectively inhibit. The long-term data from the PIONEER study showed that over a median follow-up of nearly four years, the safety of AYVAKIT remained consistent with earlier findings. The discontinuation rate due to adverse events related to the treatment was low at 3%.
