Sanofi's Blueprint Touts 4-Year AYVAKIT Data

Blueprint Medicines, a Sanofi company, announced new four-year data from its PIONEER study on the drug AYVAKIT. The results reportedly show a sustained benefit and long-term safety profile for patients with indolent systemic mastocytosis, a rare blood disorder. The company presented 12 datasets on the condition at the 2026 AAAAI Annual Meeting.

French pharmaceutical giant Sanofi completed its acquisition of Blueprint Medicines in July 2025 for approximately $9.1 billion. The deal was a strategic move to bolster Sanofi's immunology pipeline, giving it control of AYVAKIT and an established presence among immunology specialists. Indolent systemic mastocytosis (ISM) is the most common form of systemic mastocytosis, a rare disorder caused by the overproduction of abnormal mast cells. These cells can accumulate in the skin, bone marrow, and internal organs, releasing large amounts of histamine that can cause debilitating symptoms, including skin lesions, flushing, gastrointestinal distress, and bone pain. AYVAKIT (avapritinib) is a precision therapy known as a tyrosine kinase inhibitor. It is designed to specifically target and inhibit KIT D816V, a gene mutation that is the primary driver of the disease in the vast majority of patients. The U.S. Food and Drug Administration (FDA) approved AYVAKIT for adults with indolent systemic mastocytosis on May 22, 2023. This marked a significant shift in treatment, making it the first and only therapy approved to treat the underlying cause of the disease, rather than just managing its symptoms. The initial approval was based on the results from the Phase 2 PIONEER trial, which met its primary endpoint by showing a statistically significant improvement in the total symptom score for patients taking AYVAKIT compared to a placebo. The drug was previously approved in June 2021 for advanced forms of systemic mastocytosis.
