Blueprint Medicines Touts 4-Year Drug Data

Indolent systemic mastocytosis (ISM) is a rare blood disorder where the body produces too many mast cells. In about 95% of cases, the disease is driven by a specific genetic mutation known as KIT D816V. These excess mast cells can accumulate in various tissues, leading to a wide range of debilitating symptoms including skin lesions, gastrointestinal issues, bone pain, and cognitive problems like brain fog. AYVAKIT (avapritinib) is a targeted therapy designed to inhibit the KIT D816V mutation, addressing the underlying cause of the disease rather than just managing its symptoms. The U.S. Food and Drug Administration (FDA) initially approved the drug in January 2020 for a type of gastrointestinal tumor. Its approval was later expanded in May 2023 to include adults with ISM. The PIONEER trial was a pivotal Phase 2, randomized, and placebo-controlled study designed to evaluate AYVAKIT's effectiveness and safety in ISM patients whose symptoms were not well-managed by other supportive care. The study's primary endpoint measured the change in the total symptom score (TSS) as reported by patients. Key secondary endpoints included reductions in mast cell burden, which were assessed by measuring serum tryptase levels and the presence of the KIT D816V mutation in the blood. The acquisition of Blueprint Medicines by Sanofi was completed in July 2025, bringing AYVAKIT into the portfolio of the global pharmaceutical company. The deal, valued at approximately $9.1 billion, was aimed at expanding Sanofi's presence in rare immunological diseases and leveraging Blueprint's expertise in this area.
