Sanofi's AYVAKIT Shows Long-Term Safety
Four-year data from a real-world study showed that AYVAKIT, a drug for the rare disease indolent systemic mastocytosis, has a sustained benefit and long-term safety profile. The results were presented by Blueprint Medicines, a Sanofi company, at the 2026 AAAAI Annual Meeting.
Indolent systemic mastocytosis (ISM) is a chronic condition caused by an overproduction of mast cells, leading to a wide range of debilitating symptoms like skin lesions, gastrointestinal issues, and neurocognitive problems. In about 95% of cases, the disease is driven by a specific genetic mutation known as KIT D816V. AYVAKIT (avapritinib) is a targeted therapy designed to specifically inhibit this KIT D816V mutation, addressing the underlying cause of the disease rather than just managing its symptoms. The U.S. Food and Drug Administration (FDA) approved AYVAKIT for adults with indolent systemic mastocytosis in May 2023, making it the first and only approved medicine for this condition. The long-term data comes from the PIONEER clinical trial, a significant study in the field of ISM. The results presented at the 2026 American Academy of Allergy, Asthma and Immunology (AAAAI) Annual Meeting demonstrated that the benefits of AYVAKIT in controlling symptoms and improving quality of life are sustained over a four-year period. Beyond the clinical trial setting, a real-world study of AYVAKIT also highlighted improvements across various symptoms for patients receiving the therapy in a community practice setting. This reinforces the drug's effectiveness and tolerability in everyday clinical use. The development and presentation of these findings come from Blueprint Medicines, which was acquired by Sanofi. This acquisition has bolstered Sanofi's portfolio in rare immunological diseases. The safety profile of AYVAKIT remained consistent and favorable over the long-term, a crucial factor for a chronic medication. Common side effects observed in studies have included eye edema, dizziness, peripheral edema, and flushing.
