Sanofi Drug Shows Long-Term Benefit in Rare Disease
Blueprint Medicines, a Sanofi company, announced new four-year data from its PIONEER study at the 2026 AAAAI Annual Meeting. The results show that its drug AYVAKIT provides sustained benefits and has a strong long-term safety profile for patients with indolent systemic mastocytosis, a rare blood disorder.
Indolent systemic mastocytosis is a rare blood disorder where the body produces too many mast cells. These cells, a type of white blood cell, normally help the immune system but in this condition, they accumulate in organs and tissues like the skin and bone marrow. This buildup can lead to a range of symptoms including skin rashes, gastrointestinal issues, and even anaphylaxis. The disorder is most often caused by a specific genetic mutation in the KIT gene. While many patients experience mild to moderate symptoms and have a normal life expectancy, the chronic nature of the disease can significantly impact their quality of life. Indolent systemic mastocytosis accounts for the vast majority of all systemic mastocytosis cases. Prior to targeted therapies like AYVAKIT, treatment for indolent systemic mastocytosis primarily focused on managing symptoms. This often involved a combination of H1 and H2 antihistamines to address skin and gastrointestinal issues, as well as mast cell stabilizers. Patients are also advised to carry epinephrine injectors to treat severe allergic reactions. AYVAKIT (avapritinib) is a tyrosine kinase inhibitor that specifically targets the KIT D816V mutation, which is present in about 95% of systemic mastocytosis cases. The U.S. Food and Drug Administration previously approved the drug for adult patients with advanced systemic mastocytosis. The PIONEER study is evaluating its effectiveness and safety in the larger population of patients with the indolent form of the disease.
